Overview

(+)-14-hydroxymorphinone is a synthetic (+)-enantiomer of the morphinan derivative 14-hydroxymorphinone. Unlike the naturally occurring (-)-isomers of opioids, (+)-isomers do not interact with classical mu, delta, or kappa opioid receptors. Instead, (+)-14-hydroxymorphinone acts as a potent antagonist of Toll-like receptor 4 (TLR4), specifically inhibiting the TLR4-TRIF signaling pathway. Developed by researchers at the Changchun Institute of Applied Chemistry, it has demonstrated significant potential in preclinical models for treating morphine addiction by reducing behavioral sensitization and conditioned place preference. This effect is mediated through the suppression of microglial activation and the reduction of pro-inflammatory cytokines like TNF-α in key brain regions such as the medial prefrontal cortex and ventral tegmental area. It is characterized by high oral bioavailability and a favorable pharmacokinetic profile compared to other related precursors.
