**(123B9)2-L2-PTX** is a preclinical EphA2-targeting peptide-drug conjugate composed of a dimeric 123B9 EphA2-agonist peptide covalently linked through an L2 linker to the microtubule-stabilizing cytotoxic payload paclitaxel. The dimeric peptide activates and promotes oligomerization of ephrin type-A receptor 2 at nanomolar concentrations, enabling receptor-directed delivery of paclitaxel to EphA2-expressing tumor cells. In breast-cancer metastasis mouse models, the conjugate reduced circulating tumor cells and lung metastases, including superior antimetastatic activity versus albumin-bound paclitaxel.
