(4S,5E,7Z,10Z,13Z,16Z,19Z)-4-hydroxydocosa-5,7,10,13,16,19-hexaenoic acid, also known as 4-HDHA or 4-OHDHA, is a bioactive metabolite derived from the omega-3 polyunsaturated fatty acid docosahexaenoic acid (DHA). It is formed through the 5-lipoxygenase (5-LOX) enzymatic pathway. This small molecule acts as a potent agonist of peroxisome proliferator-activated receptor gamma (PPARγ), mediating antiangiogenic and anti-inflammatory effects. Research indicates that 4-HDHA directly inhibits endothelial cell proliferation and sprouting angiogenesis, suggesting its potential in treating conditions characterized by pathological neovascularization, such as proliferative retinopathy. It has also demonstrated anti-inflammatory properties by suppressing inflammatory gene expression and alleviating symptoms in models of colitis.
