(Pro3)GIP(1-16) is a C-terminally truncated peptide analog of glucose-dependent insulinotropic polypeptide (GIP) featuring a proline substitution at the third amino acid position. It was developed as a potential small molecular weight GIP receptor antagonist for the treatment of obesity and type 2 diabetes. In experimental models, including BRIN-BD11 cells and obese diabetic (ob/ob) mice, the peptide demonstrated weak antagonistic properties against native GIP. However, chronic administration in ob/ob mice failed to produce significant improvements in metabolic parameters such as glucose tolerance, insulin sensitivity, or body weight. Due to its significantly reduced biological potency compared to full-length (Pro3)GIP, researchers concluded that the molecule lacks therapeutic utility.
