(R)-DOTAP + Trp2 is an investigational combination immunotherapy consisting of the cationic lipid (R)-DOTAP and the melanoma-associated peptide antigen Trp2 (tyrosinase-related protein 2). (R)-DOTAP is the immunologically active enantiomer of DOTAP (1,2-dioleoyl-3-trimethylammonium-propane), serving as both a delivery vehicle and immune adjuvant, while Trp2 is a tumor-associated antigen. When formulated together, (R)-DOTAP enhances the uptake and immunogenicity of Trp2, breaking immune tolerance and inducing strong CD8+ T cell and CTL responses against melanoma in preclinical models. This combination shows the ability to delay tumor growth and activate tumor-infiltrating lymphocytes, making it a promising therapeutic cancer vaccine approach in melanoma and potentially other cancers[2][1].
