Overview

[11C]FLB 457 is a radiopharmaceutical positron emission tomography (PET) ligand labeled with carbon-11, developed for in vivo quantification of dopamine D2 and D3 receptors, especially in extrastriatal regions of the human brain. It has very high affinity (Ki ≈ 20 pmol/L) and selectivity for these receptor subtypes, making it useful for imaging low-density D2/D3 populations outside the striatum, such as in the thalamus, cingulate, temporal, and frontal cortices. Its binding in the brain is highly specific and reversible, and the ligand is primarily used as a research tool in neuropsychiatric studies, including those involving schizophrenia and other dopaminergic pathologies[1][3][4][6].
