[11C]FPEB is a radiopharmaceutical tracer designed for positron emission tomography (PET) imaging of the metabotropic glutamate receptor 5 (mGluR5) in the central nervous system. Chemically a derivative of the mGluR5 antagonist MPEP, it acts as a highly selective small molecule negative allosteric modulator (NAM). By binding to an allosteric site on the mGluR5 receptor, it allows for the non-invasive quantification of receptor density and distribution in the living brain. Developed primarily by the National Institute of Mental Health (NIMH) and validated by institutions such as Yale University, [11C]FPEB is a critical tool in clinical research for studying the pathophysiology of various neurological and psychiatric disorders, including Alzheimer's disease, epilepsy, Fragile X syndrome, and major depressive disorder. It is preferred over earlier tracers due to its high binding affinity, favorable brain kinetics, and low level of non-specific binding.
