[14C]PF-07054894 is a radiolabeled version of PF-07054894, a potent and selective small molecule antagonist of the C-C chemokine receptor type 2 (CCR2). Developed by Pfizer, the compound was primarily investigated for the treatment of inflammatory bowel diseases, specifically ulcerative colitis. CCR2 is a G protein-coupled receptor that plays a critical role in the recruitment of inflammatory monocytes to tissues; by antagonizing this receptor, PF-07054894 aims to attenuate chronic inflammation. The [14C]-labeled variant was specifically utilized in Phase 1 clinical trials to conduct mass balance and ADME (absorption, distribution, metabolism, and excretion) studies, allowing researchers to track the drug's metabolic pathways and elimination routes in human subjects. Although it reached Phase 2 development for ulcerative colitis, the program was subsequently discontinued by Pfizer.
