[177Lu]Lu-DOTA-RW03 is a therapeutic radiopharmaceutical consisting of a fully humanized monoclonal antibody, RW03, which targets CD133 (prominin-1), conjugated to the beta-emitting radioisotope lutetium-177 (177Lu) via the DOTA chelator. CD133 is a glycoprotein frequently expressed on cancer stem cells in various solid tumors, including glioblastoma and colorectal cancer. By binding specifically to CD133-positive cells, the agent delivers localized ionizing radiation, resulting in DNA damage and apoptosis. In preclinical studies presented at AACR 2026, Imaging Mass Cytometry (IMC) was used to assess the single-cell biodistribution and functional response of the drug, demonstrating its ability to target CD133+ tumor regions and induce therapeutic effects.
