Overview

[177Lu]Lu-DWJ155 is an investigational radioligand therapy (RLT) developed by **Novartis** for the treatment of advanced solid tumors. It consists of a targeting ligand, likely a peptide, that specifically binds to the **Human epidermal growth factor receptor 3 (HER3)**, conjugated to the therapeutic beta-emitting radioisotope **Lutetium-177 (177Lu)**. HER3 is a member of the ErbB family of receptor tyrosine kinases and is frequently overexpressed in various cancers, including breast, non-small cell lung, and gastric cancers, where it is associated with tumor progression and resistance to standard therapies. By targeting HER3, [177Lu]Lu-DWJ155 delivers localized cytotoxic radiation directly to tumor cells, causing DNA damage and apoptosis. The agent is being studied in a Phase I clinical trial (NCT06414109) as part of a theranostic pair with the PET imaging agent [68Ga]Ga-DWJ155, which allows for the selection of patients based on their tumor's HER3 expression levels.
