Drug intelligence / Profile preview
[18F]-D-AIP(FPPA) is a first-in-class, small-molecule peptidomimetic positron emission tomography (PET) tracer designed for the selective imaging of soluble amyloid-beta (Aβ) oligomers [18F]-D-AIP(FPPA). Developed by researchers at McGill University, the tracer is derived from D-AIP (D-amino acid Aβ42 oligomer-interacting Peptide), an 8-amino acid peptide that specifically binds to and neutralizes toxic Aβ42 oligomers. By substituting the C-terminus of D-AIP with a fluorine-18 labeled phenylalanine mimic ([18F]-Fppa) using a novel radiolabeling methodology, the resulting tracer maintains high affinity for Aβ oligomers while minimizing plaque-related signals [18F]-D-AIP(FPPA). This allows for the visualization of Aβ aggregation in its earliest stages (*in statu nascendi*), potentially enabling early diagnosis of Alzheimer's disease and the evaluation of oligomer-specific therapeutics [18F]-D-AIP(FPPA).
