Drug intelligence / Profile preview
[18F]asyn-44 is a fluorine-18 labeled pyridothiophene-based positron emission tomography (PET) radiotracer designed for the in vivo imaging of pathological alpha-synuclein (α-syn) aggregates. Developed by researchers at the Centre for Addiction and Mental Health (CAMH), the University of Pittsburgh, and collaborating institutions, the tracer was intended to aid in the early diagnosis, differential classification, and therapeutic monitoring of synucleinopathies, including Parkinson's disease (PD), multiple system atrophy (MSA), and dementia with Lewy bodies (DLB). Although [18F]asyn-44 demonstrated high binding affinity to α-syn aggregates in vitro and good initial brain uptake in preclinical animal models (including rodents, pigs, and non-human primates), its further clinical translation was halted due to rapid metabolism and the formation of troublesome radiometabolites.
