[18F]F-Bapi-Fab8D3FcRn is a fluorine-18 radiolabeled bispecific antibody designed for positron emission tomography (PET) imaging of amyloid-beta (Aβ) pathology in the brain, primarily for Alzheimer's disease research. It is derived from the Aβ-targeting antibody bapineuzumab (Bapi) and incorporates a fragment of the 8D3 antibody (Fab8D3), which binds to the transferrin receptor (TfR) to facilitate receptor-mediated transcytosis across the blood-brain barrier. To optimize imaging contrast, the antibody is engineered with reduced affinity for the neonatal Fc receptor (FcRn), promoting faster systemic clearance and reducing background signal in the blood. This tracer allows for same-day PET imaging of brain Aβ deposits by combining efficient brain uptake with rapid blood elimination.
