Overview

[18F]F-BapiFcRn is a radiolabeled monoclonal antibody tracer designed for positron emission tomography (PET) imaging of amyloid-beta (Aβ) pathology in Alzheimer's disease. It is based on bapineuzumab, a humanized antibody that targets the N-terminus of the Aβ peptide. The antibody has been engineered with specific mutations in its Fc region to reduce its binding affinity for the neonatal Fc receptor (FcRn). This modification facilitates rapid clearance of the tracer from the blood, thereby reducing background radioactivity and improving the signal-to-noise ratio for brain imaging. Labeled with the radioisotope fluorine-18, [18F]F-BapiFcRn is used in preclinical research to visualize Aβ plaques and to serve as a reference for evaluating the brain uptake of bispecific antibodies that utilize receptor-mediated transcytosis.
