Drug intelligence / Profile preview
[18F]F-FAPI-74 is a fluorine-18 labeled small molecule radiopharmaceutical and positron emission tomography (PET) imaging tracer that targets fibroblast activation protein alpha (FAP). FAP is a transmembrane serine protease highly overexpressed on cancer-associated fibroblasts (CAFs) in the tumor microenvironment of more than 90% of epithelial carcinomas, as well as in active fibrotic lesions. Originally developed by researchers at Heidelberg University Hospital, [18F]F-FAPI-74 is licensed to SOFIE Biosciences for clinical development and commercialization in the United States, while GE HealthCare holds the development and commercialization rights outside the United States. [18F]F-FAPI-74 is being evaluated in Phase 3 clinical trials (such as FAPI-GO and FAPI-PRO) for the detection of metastatic disease in patients with gastroesophageal cancer and pancreatic ductal adenocarcinoma (PDAC). It is also being investigated in Phase 2 trials for various other solid tumors (including breast, bladder, colorectal, and ovarian cancers) and in Phase 1 trials for imaging tissue fibrosis and sarcoidosis.
