Drug intelligence / Profile preview
`[18F]F-siPSMA-14` (also referred to as `[18F]siPSMA-14`) is a novel silicon-based radiofluorinated prostate-specific membrane antigen (PSMA)-targeted radioligand developed for positron emission tomography (PET) imaging of prostate cancer. It incorporates a silicon-fluoride acceptor (SiFA) moiety, which enables simple, rapid, and scalable radiolabeling via a one-step room-temperature fluorine-18 for fluorine-19 isotopic exchange reaction, avoiding the need for complex high-temperature synthesis or HPLC purification. Preclinically, `[18F]F-siPSMA-14` has demonstrated high binding affinity and specific accumulation in PSMA-positive tumor models. In clinical proof-of-concept studies, it has shown favorable biodistribution, moderate radiation dosimetry, and intense uptake in primary and metastatic prostate cancer lesions, enabling high-contrast PET imaging even without forced diuresis.
