Overview

[18F]PF-06445974 is a fluorine-18 labeled positron emission tomography (PET) radioligand that preferentially binds to phosphodiesterase 4B (PDE4B), an enzyme involved in the termination of cyclic adenosine monophosphate (cAMP) signaling. It was developed to enable in vivo imaging and quantification of PDE4B expression, particularly in the brain, for research into neuropsychiatric and inflammatory disorders. The compound demonstrates high selectivity for PDE4B over other PDE subtypes, excellent brain permeability, and high target specificity. Its primary use is as a biomarker for neuroinflammation and as a tool to study cAMP signaling pathways in humans and animals. The radioligand has been evaluated in first-in-human studies for its ability to quantify PDE4B binding using PET imaging[2][5][6][8].
