Overview

[212Pb]Pb-AG1002 is an investigational targeted alpha therapy (TAT) radiopharmaceutical designed for the treatment of Glypican-3 (GPC3)-positive solid tumors, specifically hepatocellular carcinoma (HCC). It consists of the alpha-emitting radioisotope Lead-212 (212Pb) conjugated to a high-affinity targeting moiety directed against GPC3, a protein frequently overexpressed in HCC but with limited expression in normal adult tissues. Lead-212 serves as a potent source of alpha radiation, which delivers high linear energy transfer (LET) to target cells, resulting in complex double-strand DNA breaks that are difficult for cells to repair, thereby inducing apoptosis. This approach aims to provide high therapeutic efficacy while minimizing off-target toxicity to healthy tissues. The drug is being developed by AlphaGen Therapeutics.
