[225Ac]Ac-DOTA-trastuzumab IgG is a targeted alpha-particle radioimmunotherapy (TART) candidate being developed by the University of Toronto for the treatment of HER2-positive breast cancer. The therapeutic agent consists of the anti-HER2 monoclonal antibody trastuzumab (IgG) conjugated via a DOTA chelator to the alpha-particle emitter actinium-225 (225Ac). By binding specifically to human epidermal growth factor receptor 2 (HER2) overexpressed on breast cancer cells, the radioconjugate delivers highly cytotoxic alpha radiation directly to the tumor site, inducing DNA double-strand breaks and tumor cell death while minimizing damage to surrounding healthy tissues.
