[225Ac]Ac-mcp-TDI-Y-010 is a targeted alpha-particle radioimmunotherapy (RIT) agent designed for the treatment of neuroendocrine cancers. It consists of the monoclonal antibody TDI-Y-010, which specifically targets Delta-like ligand 3 (DLL3), conjugated to the alpha-emitting radionuclide Actinium-225 (225Ac) using a MACROPA-derived chelator (mcp). DLL3 is an atypical Notch ligand that is highly overexpressed on the surface of various neuroendocrine tumors, such as small cell lung cancer (SCLC) and neuroendocrine prostate cancer (NEPC), while having minimal expression in normal tissues. Upon binding to DLL3-expressing cells, the agent delivers high-energy alpha particles that induce lethal double-stranded DNA breaks, leading to tumor cell death. Preclinical evaluations have shown significant antitumor activity and manageable toxicity, positioning it as a promising candidate for clinical development in DLL3-positive malignancies.
