[225Ac]Ac-SibuDAB is an experimental radiopharmaceutical designed for targeted alpha-therapy (TAT) of prostate cancer. It consists of the alpha-emitting radioisotope actinium-225 complexed with SibuDAB, a small-molecule ligand that targets Prostate-Specific Membrane Antigen (PSMA). SibuDAB is uniquely engineered with an albumin-binding moiety (derived from ibuprofen) to enhance its pharmacokinetic profile by extending its half-life in the blood. This prolonged circulation allows for greater accumulation in PSMA-expressing tumor tissues compared to conventional ligands like PSMA-617. Preclinical data indicate that [225Ac]Ac-SibuDAB provides superior therapeutic efficacy and tumor eradication in mouse models of prostate cancer, making it a promising candidate for treating metastatic disease.
