[225Ac]hu11B6-IgG1 is a targeted alpha-particle radioimmunotherapy (RIT) candidate designed for the treatment of prostate cancer. It consists of the humanized monoclonal antibody hu11B6, which specifically targets human kallikrein-related peptidase 2 (hK2), labeled with the alpha-emitting radioisotope Actinium-225 (225Ac). hK2 is a prostate-specific enzyme regulated by the androgen receptor and is highly expressed in prostate cancer cells. Upon binding to hK2, the hu11B6 antibody is internalized, delivering the potent alpha radiation directly into the tumor cells. This localized radiation induces lethal DNA double-strand breaks, leading to cell death while minimizing damage to surrounding healthy tissues. Research has investigated various IgG subclasses and fractionation schedules to optimize the therapeutic efficacy and safety profile of this agent.
