Drug intelligence / Profile preview
[227Th]Th-Lumi804-DUNP19 is a preclinical-stage alpha-emitting radioimmunotherapy (RIT) consisting of the humanized monoclonal antibody DUNP19 conjugated to the radioactive isotope thorium-227 ([227Th]) via the macrocyclic, octadentate 1,2-HOPO-based chelator Lumi804. The antibody DUNP19 targets leucine-rich repeat-containing protein 15 (LRRC15), a protein highly expressed on both cancer cells and cancer-associated fibroblasts (CAFs) in the tumor microenvironment, but negligible in healthy tissues. By delivering high-linear energy transfer (LET) alpha particles directly to LRRC15-expressing cells, the radiopharmaceutical induces DNA double-strand breaks, leading to cell death in both the tumor and its protective stromal compartment. This dual-action targeting strategy is designed to overcome therapeutic resistance in aggressive, fibrous malignancies such as glioblastoma and osteosarcoma.
