[64Cu] Macrin is a diagnostic radiopharmaceutical consisting of a 64Cu-labeled polyglucose nanoparticle designed for quantitative positron emission tomography (PET) imaging of macrophages. It selectively accumulates in macrophages with >90% specificity, allowing for non-invasive visualization and quantification of macrophage distribution in tissues. The radioactive half-life of 64Cu (12.7 hours) enables imaging at 24 hours post-injection, providing high signal-to-noise imaging of macrophage uptake. This makes it particularly useful for detecting inflammatory "hotspots" and quantifying local macrophage density in various conditions. [64Cu] Macrin differs from previous cross-linked dextran nanoparticles in that all components are biodegradable and safe, with L-lysine used as the crosslinker. Its biological properties are optimized to avoid rapid renal clearance and maximize accumulation in macrophages within solid tissues and tumors.
