Drug intelligence / Profile preview
[64Cu]Cu-FMPA is a tumor-targeted, photoacid-responsive copper-based metal-organic framework (MOF) nanoplatform radiolabeled with copper-64 ([64Cu]) and encapsulating a p53 agonist. Developed by researchers at the Huazhong University of Science and Technology, it is designed as a theranostic agent for triple-negative breast cancer (TNBC). The platform integrates positron emission tomography (PET) imaging with radionuclide therapy and cuproptosis induction. Upon localized light irradiation, photoacid generation triggers the degradation of the MOF, enabling spatiotemporally controlled release of copper and the encapsulated p53 agonist. The p53 agonist suppresses glycolysis and induces mitochondrial dysfunction, which sensitizes tumor cells to copper-induced cell death (cuproptosis). Concurrently, the [64Cu] isotope provides high-contrast PET imaging and localized cytotoxic radiation.
