Drug intelligence / Profile preview
[64Cu]Cu-HTE1PA-9E7.4 is a preclinical theranostic radioimmunoconjugate consisting of the rat anti-mouse CD138 (syndecan-1) monoclonal antibody (clone 9E7.4) conjugated to the monopicolinate cyclam-based chelator HTE1PA (TE1PA) and radiolabeled with the positron- and beta-emitting radionuclide copper-64 (64Cu). Developed by researchers at Nantes Université, CNRS, Inserm, and the University of Brest, this agent is designed for immuno-PET imaging and targeted radionuclide therapy (TRT) of multiple myeloma. The 9E7.4 antibody specifically targets CD138, a cell surface proteoglycan highly overexpressed on multiple myeloma cells, while the HTE1PA chelator provides high thermodynamic stability and kinetic inertness, preventing transchelation of copper-64 in vivo. Preclinical studies in murine multiple myeloma models have demonstrated high tumor uptake, excellent PET imaging contrast, and therapeutic efficacy in delaying tumor growth and extending survival.
