[64Cu]Cu-NOTA-EV-F(ab')2 is a copper-64 (^64Cu)-labeled radiopharmaceutical probe constructed from the F(ab')2 fragment of enfortumab vedotin (EV), an antibody-drug conjugate directed against Nectin-4, conjugated to the chelator NOTA for radiolabeling. This agent is used for immunoPET imaging to visualize Nectin-4-expressing tumors, primarily in models of gastric cancer (GC) and non-small cell lung cancer (NSCLC). The F(ab')2 fragment is generated enzymatically from EV, conjugated to NOTA, and subsequently radiolabeled with copper-64. The resulting probe demonstrates rapid, specific, and high-contrast tumor imaging, with rapid tumor uptake and faster blood clearance compared to the full-length antibody. Mechanistically, the agent binds specifically to Nectin-4, as confirmed by blocking studies. Its main purpose is for noninvasive PET-based tumor imaging and characterization, serving as a molecular imaging tool rather than a therapeutic.[1][3][2][6][8]
