Overview

[64Cu]Cu-NOTA-TP is a radiopharmaceutical conjugate consisting of a cytotoxic terpyridine-platinum (TP) complex and a NOTA (1,4,7-triazacyclononane-1,4,7-triacetic acid) chelator labeled with the radioisotope copper-64 (64Cu). The terpyridine-platinum moiety functions as a potent DNA-intercalating agent that exerts selective cytotoxicity against cancer cells. Labeled with copper-64, the molecule acts as a radiotheranostic agent, enabling both PET imaging via positron emission and potential radiotherapy. In preclinical research, specifically concerning prostate cancer, [64Cu]Cu-NOTA-TP is primarily utilized as a monomeric control or foundational scaffold to evaluate the efficacy and targeting specificity of more complex agents, such as the PSMA-targeted derivative [64Cu]Cu-NOTA-TP-PSMA. Its mechanism involves cellular uptake followed by nuclear localization where the platinum complex interacts with genomic DNA.
