[68Ga]-DOTA-Z1085 is a radiopharmaceutical PET imaging tracer designed to target the pro-inflammatory cytokine tumour necrosis factor (TNF). It consists of a small engineered affibody molecule, Z1085, which has high binding affinity for TNF, conjugated to the chelator DOTA and radiolabeled with Gallium-68. The tracer is being developed for the non-invasive assessment of TNF distribution and availability in patients with Rheumatoid Arthritis (RA), potentially allowing for patient stratification and treatment monitoring. Preclinical studies in rat models of arthritis have demonstrated its ability to accumulate in inflamed joints with a high signal-to-background ratio, correlating with synovial inflammation and macrophage infiltration.
