Overview

[68Ga]Ga-DOTA-(2P-PEG4)2 is a gallium-68 labeled radiopharmaceutical designed for the positron emission tomography (PET) imaging of prostate cancer. It is a multivalent molecular probe consisting of a PSMA-targeting dimer conjugated to polyethylene glycol (PEG4) chains and a DOTA chelator. The design leverages the multivalent effect to enhance binding affinity to the prostate-specific membrane antigen (PSMA) and utilizes PEGylation to optimize pharmacokinetics, specifically aiming for higher tumor accumulation and rapid renal clearance. In preclinical studies, it demonstrated superior tumor-to-organ ratios compared to monomeric counterparts, making it a promising candidate for the diagnostic localization of PSMA-positive lesions.
