[68Ga]Ga-NOTA-ABDG2 is a gallium-68 labeled radiopharmaceutical developed for the immuno-positron emission tomography (immunoPET) imaging of hepatocellular carcinoma (HCC). The agent targets Glypican 3 (GPC3), a cell-surface proteoglycan that is highly and specifically expressed in HCC tissues. Structurally, the probe consists of a GPC3-specific single-domain antibody (sdAb), known as G2, which is fused to an albumin-binding domain (ABD) to optimize its pharmacokinetic profile. The ABD fusion serves to extend the circulatory half-life and reduce renal uptake, addressing common limitations of small antibody fragments. The protein is conjugated to the chelator NOTA to facilitate labeling with the positron-emitting isotope Gallium-68. Preclinical studies indicate that [68Ga]Ga-NOTA-ABDG2 provides high-contrast imaging of GPC3-positive tumors, making it a potential candidate for the diagnosis and staging of HCC.
