[68Ga]Ga-NOTA-anti-CD70 VHH is a preclinical radiopharmaceutical imaging agent consisting of a single-domain antibody fragment (VHH or nanobody) targeting the CD70 antigen, site-specifically conjugated to a NOTA (1,4,7-triazacyclononane-1,4,7-triacetic acid) chelator and labeled with the positron-emitting isotope Gallium-68 (68Ga). CD70 is a costimulatory ligand in the tumor necrosis factor (TNF) superfamily that is constitutively expressed in various solid tumors and hematologic malignancies, such as renal cell carcinoma and acute myeloid leukemia, while maintaining limited expression in normal tissues. This tracer is designed for non-invasive positron emission tomography (PET) imaging to identify patients with CD70-positive tumors and to monitor CD70 expression levels longitudinally during treatment with anti-CD70 therapies, such as monoclonal antibodies or CAR-T cells. The VHH fragment was engineered with a C-terminal C-Direct tag to allow for site-specific conjugation, ensuring high binding affinity and a homogeneous product suitable for clinical translation.
