[68Ga]Ga-PSMA-D5 is a radiopharmaceutical diagnostic agent designed for PET imaging of prostate-specific membrane antigen (PSMA) positive lesions in men with prostate cancer. It contains a DOTA chelator that binds to gallium-68, creating a radioactive tracer that targets PSMA, which is overexpressed in prostate cancer cells. In preclinical studies, [68Ga]Ga-PSMA-D5 demonstrated higher tumor uptake (SUVmax = 3.51) and lower renal uptake (T/K = 1.84) compared to [68Ga]Ga-PSMA-11. The advantages of [68Ga]Ga-PSMA-D5 include high tumor uptake, straightforward synthesis, and convenient labeling. Additionally, its DOTA chelator allows for potential labeling with therapeutic radionuclides such as 177Lu and 225Ac, providing possibilities for targeted radioligand therapy in prostate cancer.
