Drug intelligence / Profile preview
[89Zr]Zr-DFO*-NCS-Abeta-mAb-scFab8D3 (also known as [89Zr]Zr-DFO*-NCS-Adu-8D3) is a preclinical radiolabeled bispecific monoclonal antibody conjugate developed by Amsterdam UMC and Lundbeck for positron emission tomography (PET) imaging of amyloid-beta (Aβ) pathology in Alzheimer's disease. The agent consists of an aducanumab-derived antibody targeting Aβ, fused to a single-chain Fab fragment of the 8D3 antibody targeting transferrin receptor 1 (TfR1) to facilitate receptor-mediated transcytosis across the blood-brain barrier (BBB). It is conjugated with the novel bifunctional chelator DFO*-NCS and radiolabeled with the positron emitter zirconium-89 (89Zr). Preclinical studies have demonstrated that the use of the DFO* chelator provides superior stability and high, specific brain uptake, enabling the early detection of Aβ plaque pathology and the quantification of antibody brain delivery.
