[99mTc]-TcL6 is an investigational radiopharmaceutical imaging agent designed for the detection and visualization of tumors using Single-Photon Emission Computed Tomography (SPECT). The agent consists of a technetium-99m tricarbonyl core coordinated to a specific ligand, designated L6, which incorporates a D-histidine amino acid moiety as a pendant group. This design leverages the metabolic alterations characteristic of malignant cells, specifically the upregulation of amino acid transporters required for rapid tumor growth. By mimicking natural metabolic substrates, [99mTc]-TcL6 achieves targeted accumulation and internalization within tumor tissues, such as lung and breast cancer. Preclinical studies in A549 lung cancer xenograft models have demonstrated significant cellular uptake and favorable tumor-to-nontarget organ ratios, suggesting its potential as a sensitive tool for oncological imaging and metabolic assessment.
