[99mTc]HYNIC-cRGDfk-NPY is a novel radiolabeled heterodimer peptide dual-receptor-targeting probe designed for SPECT/CT imaging of breast tumors. It consists of a cyclic RGD peptide motif (cRGDfk) that targets tumor angiogenesis by binding to integrin αvβ3, and a neuropeptide Y (NPY) analog motif that targets neuropeptide Y receptor type 1 (Y1R), which is highly overexpressed in breast cancers. The peptide is conjugated to 6-hydrazinonicotinamide (HYNIC) as a chelator and radiolabeled with technetium-99m (99mTc). Preclinical studies in mouse models of estrogen receptor-positive (MCF-7) and triple-negative (MDA-MB-231) breast cancer have demonstrated rapid blood clearance, renal excretion, and high tumor uptake, highlighting its potential as a diagnostic imaging agent.
