[99mTc]HYNIC-siRNA 1489 is a technetium-99m labeled small interfering RNA (siRNA) molecular imaging probe designed for single-photon emission computed tomography (SPECT) imaging of murine double minute 2 (MDM2) expression. MDM2 is an oncogene that plays a critical role in tumorigenesis, metastasis, and chemotherapy resistance by acting as a negative regulator of the p53 tumor suppressor. The probe consists of a specific siRNA sequence (siRNA 1489) targeting MDM2 mRNA, conjugated to the chelator hydrazinonicotinamide (HYNIC) and radiolabeled with technetium-99m ([99mTc]). Developed by researchers at Harbin Medical University, the probe is designed to evaluate MDM2 expression levels in vivo, potentially offering a non-invasive strategy for the early diagnosis and stratification of breast cancer and other MDM2-overexpressing malignancies.
