Overview

[TYR34]bPTH-(7-34) is a synthetic peptide fragment of bovine parathyroid hormone (bPTH) spanning residues 7 to 34, with a tyrosine substitution at position 34. It functions as a specific and competitive antagonist of the Parathyroid Hormone Receptor 1 (PTHR1), blocking the signaling pathways typically activated by parathyroid hormone (PTH) and parathyroid hormone-related protein (PTHrP). In preclinical research, particularly in the study of multiple myeloma and bone metabolism, this peptide is used as a pharmacological tool to investigate the role of the PTH/PTHR1 axis in disease progression and therapeutic response. Research has demonstrated that [TYR34]bPTH-(7-34) can abrogate the bone-anabolic and anti-myeloma effects induced by proteasome inhibitors like bortezomib, indicating that the PTHR1 pathway is a critical mediator of the osteoblastic response in the myeloma bone marrow microenvironment.
