Overview

α-gal(-) PDAC-ly is an experimental cancer vaccine component consisting of unprocessed pancreatic ductal adenocarcinoma (PDAC) tumor lysate. It is derived from homogenized tumor tissue resected from patients and contains a broad array of tumor-associated antigens (TAAs). In research settings, particularly those investigating α-gal epitope remodeling, α-gal(-) PDAC-ly serves as the unmodified control to evaluate the efficacy of α-gal-expressing vaccines. While it contains relevant antigens such as MUC1 and mesothelin, it lacks the α-gal epitopes (Galα1-3Galβ1-4GlcNAc-R) that facilitate enhanced uptake by antigen-presenting cells via natural anti-Gal antibodies. Studies have shown that while it can elicit some immune response, its efficacy in extending survival in animal models is significantly lower than that of its α-gal-modified counterpart.
