0166+NBP is a preclinical combination therapy being developed by AlaMab Therapeutics (a subsidiary of CSPC Pharmaceutical Group) for the treatment of stroke. The combination consists of ALMB-0166, a humanized monoclonal antibody antagonist targeting Connexin43 (Cx43) hemichannels, and NBP (butylphthalide or 3-n-butylphthalide), a small molecule neuroprotective agent. ALMB-0166 works by blocking Connexin43 hemichannels on astrocytes, thereby preventing the release of neurotoxic molecules such as ATP and glutamate, reducing neuroinflammation and glial scar formation. NBP provides complementary neuroprotective effects by reconstructing microcirculation, protecting mitochondrial function, and inhibiting neuronal apoptosis and oxidative stress. Together, the combination aims to synergistically alleviate ischemic brain injury and improve neurological recovery following a stroke.
