Overview

DACCP (1,2-diaminocyclohexane(4-carboxyphthalato)platinum(II)) is a second-generation platinum-based antineoplastic agent and an analogue of cisplatin. It was developed to overcome the limitations of cisplatin, specifically its severe nephrotoxicity and the development of drug resistance. DACCP incorporates a 1,2-diaminocyclohexane (DACH) carrier ligand, a structural feature shared with oxaliplatin, which was intended to circumvent cisplatin-resistance mechanisms. The 4-carboxyphthalato leaving group was selected to enhance aqueous solubility and reduce renal toxicity. Clinical investigations in the late 1970s and early 1980s, including Phase II trials for advanced colorectal carcinoma and malignant lymphomas, demonstrated that while DACCP was better tolerated than cisplatin—showing milder gastrointestinal side effects and no significant renal dysfunction—it lacked sufficient clinical efficacy as a single agent. Consequently, its development was discontinued in favor of other DACH-platinum derivatives.
