1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)] (DSPE-mPEG) is a PEGylated phospholipid widely utilized as a functional excipient in the development of advanced drug delivery systems, such as liposomes, micelles, and nanostructured lipid carriers. It consists of a hydrophobic distearoylphosphatidylethanolamine (DSPE) anchor and a hydrophilic methoxy-polyethylene glycol (mPEG) headgroup. When incorporated into lipid bilayers, the PEG chains extend outward to create a hydrophilic steric barrier. This barrier effectively inhibits opsonization and subsequent recognition by the mononuclear phagocyte system (MPS), thereby significantly extending the systemic circulation half-life of encapsulated therapeutic agents (the "stealth" effect). DSPE-mPEG is a critical component of several FDA-approved liposomal drugs, including liposomal doxorubicin (Doxil) and liposomal irinotecan (Onivyde), where it facilitates improved tumor accumulation via the enhanced permeability and retention (EPR) effect.
