1,2-Octanediol is a small-molecule surfactant developed by Thornton & Ross for the treatment and prevention of head louse infestation (*Pediculus humanus capitis*). It is the active ingredient in the Hedrin line of pediculicides, including a 5% formulation for active treatment and a 1% formulation for prevention. The compound acts through a physical mechanism by disrupting the cuticular lipid layer of the louse, which leads to rapid dehydration and death. Because its mode of action is physical rather than neurotoxic, it is effective against lice populations that have developed resistance to traditional chemical insecticides. Clinical studies, such as the 'Can we prevent head lice?' trial, have demonstrated its efficacy in both eradicating established infestations and preventing re-infestation when used regularly.
