Overview

C4-dietary oil (1,3-dibutyryl-2-oleoylglycerol) is a synthetic structured triacylglycerol designed to stimulate the release of incretin hormones while providing fewer calories than conventional dietary fats. It is composed of an oleic acid moiety at the sn-2 position and butyric acid at the sn-1 and sn-3 positions. Upon ingestion and subsequent digestion by gastric and pancreatic lipases, the butyric acid is cleaved, releasing 2-oleoylglycerol (2-OG). 2-OG acts as a potent agonist for the G protein-coupled receptor 119 (GPR119) located on intestinal L-cells and K-cells, triggering the secretion of glucagon-like peptide 1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). This mechanism enhances postprandial incretin responses, which can improve glucose metabolism and insulin secretion. It has been investigated as a potential dietary intervention for managing overweight individuals with type 2 diabetes and obesity.
