Overview

1β-amino-2β,25-dihydroxy-19-nor-vitamin D3 is a synthetic analog of the active form of vitamin D3, calcitriol (1α,25-dihydroxyvitamin D3). It is characterized by the replacement of the 1α-hydroxyl group with a 1β-amino group, the addition of a 2β-hydroxyl group, and the removal of the C19 methylene group (19-nor modification). This compound was developed as part of a chemical series to investigate the structure-activity relationships of A-ring modified vitamin D derivatives, specifically focusing on how amino substitutions affect biological activity. Experimental evaluation has shown that this specific isomer possesses significantly lower binding affinity for the vitamin D receptor (VDR) and the human vitamin D binding protein (hDBP) compared to the natural hormone. Despite its low receptor affinity, it has demonstrated antiproliferative activity in cancer cell lines such as MCF-7 (breast cancer) and HL-60 (leukemia) at micromolar concentrations, although it is less potent than other analogs in the same series.
