1-(5-((6-nitroquinazolin-4-yl)thio)-1,3,4-thiadiazol-2-yl)-3-(p-tolyl)urea is a novel synthetic small molecule quinazoline derivative investigated for its potential as an anticancer agent. It belongs to a series of 1,3,4-thiadiazole-aryl urea quinazolines designed to target receptor tyrosine kinases. In preclinical studies, the compound demonstrated significant antiproliferative activity against several human cancer cell lines, including A549 (lung cancer), MCF7 (hormone-dependent breast cancer), and MDA-MB231 (triple-negative breast cancer). Molecular docking and computational studies suggest that its mechanism of action involves the inhibition of Vascular Endothelial Growth Factor Receptor 2 (VEGFR-2), thereby potentially inhibiting angiogenesis and tumor cell proliferation. The compound was developed by researchers at Hacettepe University as part of an effort to identify new VEGFR-2 inhibitors with improved cytotoxic profiles.
