Overview

ACPD (1-aminocyclopentane-1,3-dicarboxylic acid) is a small molecule that serves as a broad-spectrum agonist of metabotropic glutamate receptors (mGluRs). While primarily recognized in neuroscience as a pharmacological tool for studying excitatory neurotransmission, research from the University of South Florida (Acevedo-Duncan lab) has identified ACPD as an inhibitor of atypical protein kinase C (aPKC) isoforms, specifically PKC-zeta (ζ) and PKC-iota (ι). In preclinical studies, ACPD has demonstrated the ability to decrease cell proliferation and potentially inhibit cell motility in metastasized melanoma cell lines, such as SK-MEL-2, without affecting normal melanocytes. Its role as an aPKC inhibitor suggests potential applications in oncology, particularly for cancers driven by atypical PKC signaling, such as malignant melanoma.
