1-methyl-N-propenyl-1,2,3,4-tetrahydroisoquinoline (1-Me-N-propenyl-TIQ) is a synthetic derivative of the endogenous neuroprotective agent 1-methyl-1,2,3,4-tetrahydroisoquinoline (1-MeTIQ). It has been investigated for its potential therapeutic effects in Parkinson's disease models, specifically for its ability to alleviate motor symptoms. In preclinical studies using MPTP-treated mice, 1-Me-N-propenyl-TIQ demonstrated a significant reduction in bradykinesia. However, unlike its N-propargyl counterpart, it failed to provide significant neuroprotection against the loss of striatal dopamine, nigral tyrosine hydroxylase-positive neurons, or the downregulation of the dopamine transporter (DAT). This suggests that while the propenyl modification confers some symptomatic benefit, it does not share the potent neuroprotective profile of the alkynyl-substituted derivatives in this chemical series.
