The 10-valent pneumococcal + Haemophilus influenzae protein D conjugate vaccine (commercially known as Synflorix, developed by GSK) is a pediatric vaccine designed to protect against invasive disease, pneumonia, and acute otitis media caused by Streptococcus pneumoniae and non-typeable Haemophilus influenzae (NTHi). The vaccine contains capsular polysaccharides from 10 pneumococcal serotypes (1, 4, 5, 6B, 7F, 9V, 14, 18C, 19F, and 23F), with eight conjugated to NTHi protein D as a carrier protein, and the remaining two conjugated to tetanus toxoid or diphtheria toxoid. In addition to its standard pediatric indications, the vaccine has been evaluated in clinical trials (such as the Phase 4 CHiRRP trial in Australia) to determine its efficacy in reducing respiratory exacerbations in children with chronic suppurative lung disease (CSLD), bronchiectasis, and recurrent protracted bacterial bronchitis (PBB).
