This is a combination therapy consisting of two agents: - **125I-mAb 425** is a radioimmunotherapy agent composed of a murine monoclonal antibody (mAb 425) directed against the epidermal growth factor receptor (EGFR), labeled with radioactive iodine-125. It targets EGFR-expressing tumor cells and delivers localized radiation to induce cytotoxicity. - **Temozolomide** is an oral alkylating agent that methylates DNA at the O6 and N7 positions of guanine residues, leading to DNA damage and apoptosis in rapidly dividing tumor cells. The combination has been studied as adjuvant therapy for newly diagnosed glioblastoma multiforme (GBM), particularly after surgery and radiotherapy. In clinical studies, this regimen demonstrated improved median survival compared to either agent alone or historical controls[1][3]. The mechanism leverages both targeted radiopharmaceutical action against EGFR-positive cells and systemic cytotoxic chemotherapy.
